Abstract

Abnormalities of 5HT2A receptor and receptor-linked phosphoinositide signaling (PI) have been implicated in the pathophysiology of suicide. To examine if abnormalities of 5HT2A receptors and an abnormal PI signaling system may be associated with the adolescent suicide, we determined gene expression of 5HT2A receptors, PKC, PKA, CREB, BDNF and TrkB receptors in the PFC and hippocampus of adolescent suicide victims and controls. We found that the gene expression of 5HT2A receptors was increased and of PKC alpha, beta and gamma was decreased in the PFC and hippocampus of adolescent suicide victims. We also found that the mRNA expression of PKA-R1 alpha and R1 beta, as well as CREB and BDNF, was decreased in the PFC but not the hippocampus of adolescent suicide victims. Since the physiological effects of BDNF are mediated by TrkB, we also determined mRNA for TrkB receptors. We found that the gene expression of TrkB was significantly reduced in the PFC of teenage suicide victims. These results thus suggested that an abnormality in signal transduction mechanisms may be associated with pathophysiology of teenage suicide and that this alteration in signaling system causes abnormal gene expression of the transcription factor CREB as well as its target gene BDNF.
