Abstract

In order to investigate possible pharmacodynamic interactions the following treatments were compared using an intraindividual design: a) 75 mg of the antidepressant Nomifensine, b) ca. 750 ml wine + Nomifensine Placebo, c) ca. 750 ml wine + 75 mg Nomifensine. All treatments were given as single administrations. For the treatments b) and c) the comparison was doubleblind. The effects of the different treatments were evaluated using a test battery which consisted of a number of physiological variables (pulse, blood pressure and body sway) together with a further test series to assess reaction time, ability to concentrate, visuo motor coordination, visual perception threshold and subjective mood state. Of the 21 variables, 14 showed statistically significant differences between the treatments. There was a consistent reduction in performance under both alcohol and alcohol + Nomifensine when compared to Nomifensine alone. The two situations under alcohol failed to show significant differences between one another but the decrease in performance under alcohol alone was more marked than under alcohol + Nomifensine. As far as the experimental design allows a generalisation, it may be concluded that there is no pharmacodynamic interaction between Nomifensine and alcohol especially regarding a possible potentiation of the alcoholic effects. Furthermore there were no signs of a pharmacokinetic interaction.
