Abstract

An automated ultra micro distillation technique for enzymatic determination of ethanol in blood and urine was developed, using an ADH technique after the distillation step. The technique, adapted for a Technicon Auto Analyser, allowed the determination of ethanol in concentrations down to 0.001 mg ethanol per gram blood (0.0217 mmol/l) in 1-10 (mu)l samples, diluted to 1.00 ml with sodium fluoride and run at a rate of 50-60 samples per hour. ADH interfering substances such as pyrazole, metronidazole or heavy metals did not influence the reaction. The technique is also adaptable for the determination of methanol and acetaldehyde, ammonia and phenols, and for extraction of non volatile substances.
