TY - JOUR PY - 1991// TI - Specific site distribution of TN 5 in pseudomonas putida PpGI-MI using suicide plasmid pSUP 2021 JO - Egypt. J. Microbiol. A1 - T.m.a., El Kawokgy SP - 443 EP - 451 VL - 26 IS - 3 N2 - Transposon Tn 5, which confers kanamycin-neomycin resistance, was introduced into the genome of Pseudomonas putida PpG1-M1 by conjugation with Escherichia coli SM10 containing the suicide plasmid vector pSUP2021. Approximately, 1.82% of over 2200 neomycin resistant [Nmr] colonies were mutated to auxotrophy. Most auxotrophs reverted at low frequencies [<10-8] and most prototrophic revertants were neomycin sensitive. The Tn 5-induced auxotrophic mutations were mainly distributed among genes of aspartic acid group indicating specific site-distribution of Tn 5 among the genome of Pseudomonas putida PpG1-M1
